Research Abstract

Dnmt1-associated protein 1(Dmap1)はBmi1とも会合し、Bmi1依存性の遺伝子発現抑制に関与する分子である。一方、Dmap1はNuA4 histone acetyltransferase(HAT)複合体の構成分子であることが報告されており、その機能は明らかになっていない。今回われわれはDmap1がDNA損傷修復反応の後期に必須な分子であることを見出した。マウス胎仔線維芽細胞を放射線照射すると、DNA二重鎖損傷部位にDmap1がγ-H2AXやリン酸化ATMとともに集積することが観察された。Dmap1をレンチウイルスshRNAを用いてマウス胎仔線維芽細胞でノックダウンすると自然発症的にまたレンチウイルス感染刺激によってDNA二重鎖損傷が誘導され、細胞はp53依存性のチェックポイント機能により細胞周期が停止した。重要なことにDmap1をノックダウンすると、放射線誘導性DNA二重鎖損傷部位のDNA損傷修復反応後期においてPCNAやCAF-1といったクロマチン再構成分子の集積が著明に障害されていた。生化学的解析においてもDmap1はPCNAと直接的に会合し、DNA二重鎖損傷部位にPCNAをリクルートすることが確認された。さらに、Dmap1ノックダウンによるマウス胎仔線維芽細胞の細胞周期停止はp53ノックアウトバックグラウンドにおいては回避され細胞は増殖し続けることが可能であったが、染色体異数性や分配以上が著明であり、免疫不全マウス皮下に移植した胎仔線維芽細胞は腫瘍を形成した。以上より、Dmap1はゲノム安定性の維持に必須な分子であり、癌抑制遺伝子として機能する可能性が示唆された。
