Research Abstract

ヒストンメチル化酵素の機能を解明すべく、G9a複合体の機能解析も行った。その結果、G9aとGLPが分子量の比率が野生型の細胞では1:1の複合体として存在していることを見出した。その結合様式はG9a/GLP共にC末のSET domainに依存的であることも分かった。以上のことから、G9aはGLPとの複合体として存在するときに限って生体内でヒストンのメチル化酵素として機能しうる可能性が強く示唆された。さらに詳細に解析を進めたところ、G9aの酵素活性が生体内でのメチル化に重要であり、反してGLPの活性は無くても良いことがわかった。また、GLPはG9aのタンパク質安定化に寄与する分子であることも分った。
これらの知見に加え、G9a/GLP複合体にはヒストンメチル化非依存的に転写を抑制する機能があることを見出した。さらにこの抑制能は、DNAのメチル化を呼び込むことで引き起こされることをも明らかにした。現在この知見を査読のある海外の専門誌に投稿中である。
