Research Abstract

熱ショック転写因子(HSFs)は、高等動物においてHSF1、HSF2、そしてHSF4の少なくとも3つ存在している。HSF群は、熱ショック蛋白質遺伝子の上流に存在する熱ショックエレメント(HSE)を認識し転写を制御し、そのHSE配列はnGAAn(nは任意の塩基)を1ユニットとして3つ以上の逆向きの繰り返し配列から構成されている。HSF1は熱ショックにより三量体に転換してDNA結合能を持つことがわかっているが、HSF4は三量体抑制ドメインを持たず、恒常的に三量体を形成する。最近、HSF群が感覚器の形成過程において遺伝子発現を拮抗的あるいは相補的に働くことが分かってきた。その協調的作用の分子機構を明らかにするために、HSF4のDNA結合領域を同定し、結合の特徴を明らかにした。
まず、レンズ上皮細胞を用いてクロマチン免疫沈降法を行い、71個のHSF4結合領域を見いだした。これら結合領域には、ランダム配列で同定した結合配列nGnnnの繰り返し配列が存在した。このうち58個の領域は、生後2日目のレンズを用いたクロマチン免疫沈降法でHSF4の結合が見られ、またHSF群の中でHSF4のみが結合する領域があることがわかった。次に、リコンビナントHSF4とランダムな配列のオリゴヌクレオチドを用いて、HSF4のDNA結合配列を同定した。このHSF4のDNA結合配列のすべてにnGAAnのG塩基は保存されていたが、A塩基はほとんど一致しなかった。HSF4は結合配列として同定したnGnnn配列のほとんどでHSF1やHSF2よりも強い結合を示した。以上のことから、HSF4はHSF1やHSF2に比べ、典型的HSEよりも曖昧なnGnnnに強い親和性があることが示唆された。
