植物MATE型2次代謝産物トランスポーターの機能決定

Research Abstract

ヒトのMATE(multidrug toxic compounds extrusion)型トランスポーターを発見した。このトランスポーターは腎臓や肝臓における薬物排出の最終段階を司っていることが明らかとなった。類似のトランスポーターは細菌や植物に広く存在している。植物においてはアルカロイドやフェノールなどのトランスポーターである可能性が遣伝学的な解析から示唆されているが、cDNA産物の輸送機能が測定できないために最終的な結論が得られていない。本研究は植物のリコンビナントMATE型トランスポーターの輸送機能を定量的に測定する実験系を構築し、その輸送基質を決定することを目的とした。また、この方法論を他のトランスポーターに応用することも目的とした。本年は最終年度にあたり、成果を纏めた。以下に要約する。
(1)大腸菌のプロトンポンプとトランスポーターを用いた簡便かつ高感度の再構成系を確立した。
(2)タバコ葉よりジャスモン酸により発現が誘導される遺伝子を単離、そのうち一種を昆虫細胞内で大量発現させ、精製再構成により輸送機能を測定した。予想通り、このトランスポーターは有機カチオンや両親媒性イオンを輸送した。しかも、タバコの液胞に局在しており、ニコチンの濃縮に関与する証拠を得た。
(3)ほ乳類のMATEトランスポーターが3つのサブクラスに分類できることを分子生物学的に生化学的に明らかにした。
(4)強力な阻害剤。開発した。
(5)この方法を広く植物・動物のトランスポーターに応用した。その結果、新しいトランスポーターを同定した。その例の一つはvesicular nucleotide transporter=(VNUT)である。この遺伝子は動物から植物に広く分布している。プリン性化学伝達が極めて広く分布・機能していることを示唆している。
