Research Abstract

細胞外プロトン濃度即ちpHの低下を感知して細胞機能が変化することは様々な生体調節系で観察されているが、その分子機構は不明である。最近、我々を含む国内外のグループによってOGR1ファミリーG蛋白質共役受容体が細胞外pH低下に応答して細胞内シグナル伝達系を活性化することが明らかにされた。本研究ではプロトン感受性G蛋白共役受容体の機能発現を保証するG蛋白質シグナルの特定、生体における関与を調べるために、プロトン感受性GPCR欠失マウスの作成を試みた。(1)プロトン感知性GPCRにおけるG蛋白質シグナルの特定:プロトン感知性受容体を過剰発現すると、リガンド非依存性応答として、各種細胞内転写活性(SREなど)促進、ERK活性化能の抑制など様々な作用が報告されている。そこで、プロトン感知性GPCR(OGR1、GPR4、TDAG、8G2A)を過剰発現したHEK293細胞などを用い、SREプロモーター活性、ERK活性、cAMP産生、イノシトールリン酸産生などを解析した。その結果、従来リガンド非依存性といわれていた応答が細胞外のわずかなpH変化に基づくこと、また、各プロトン感知性受容体はそれぞれ異なった組み合わせの3量体G蛋白質、低分子G蛋白質と連関し、それぞれが特徴ある細胞内シグナル系を制御していることが明らかにされた。(2)好中球の感染防御系におけるTDAG8の役割:好中球など血球系の細胞ではTDAG8が発現している。細胞外pHの低下は好中球の感染防御に重要なスーパーオキシド産生を抑制する。この作用はPKAの阻害薬H89で抑制された。実際、細胞外pH低下はcAMP産生を増加した。非特異的なOGR1受容体ファミリーのアンタゴニスであるサイコシンも細胞外pH低下によるcAMP産生作用、スーパーオキシド産生抑制作用を抑制した。このようにTDAG8/Gs/cAMP系を介していることが推定された。現在、TDAG8欠損マウス由来の好中球を用い解析している。(3)OGR1欠損マウスの作成:OGR1受容体、GPR4受容体欠損マウスの作成に成功した。現在、解析をすすめている。
