Research Abstract

これまでの研究により、細胞間接着形成において重要な役割を担う低分子量Gタンパク質Cdc42、Racの活性化には、細胞間接着分子ネクチンと細胞-基質間接着分子インテグリンαvβ3との協調的な作用が必要であることが明らかになっている。さらに詳細な検討により、そのシグナル伝達機構としてインテグリンαvβ3下流のPKC-FAKの活性化が重要であった(Ozaki, et. al. Genes Cells.2007)。また、細胞間接着形成後もインテグリンαvβ3はネクチンと細胞間接着部位で共局在するものの、インテグリンαvβ3は活性化状態から不活性化状態へと変化し、それに伴ってCdc42やRacも不活性化される。今まで、細胞間接着形成後にインテグリンav(33が不活性化される分子機構については全く不明であったが、本年度の研究により、インテグリンαvβ3の活性化に関わるI型ボスファチジルイノシトール5-キナーゼの活性が、ネクチン同士の結合によって活性化されるホスファターゼPTPμによって抑制されるためであることを明らかにした(Sakamoto et a1. J Biol Chem.2008)。インテグリンαvβ3の不活性化により細胞の運動・増殖は抑制されることから、この分子機構は細胞間接着完成後の細胞間接着維持および細胞の接触阻害機構を理解する上で極めて重要である。さらに、別の低分子量Gタンパク質Raplがネクチン-1の下流で、表皮の分化に大きな役割を担っているロリクリンの発現制御に関わっていることを見出した(Wakamatsu, et. al.J Biol Chem.2007)。ロリクリンの発現量低下により、物理的刺激に対する表皮細胞の抵抗性は著しく低下することから、ネクチン-1によるRaplの活性化は表皮細胞間の接着を維持する上で重要である。
