Research Abstract

新奇G蛋白質セプチンSept8などをモデルとして性状・機能解析を行い,以下に示す知見を得た。1.Sept8が,幼若海馬神経細胞の突起および成熟神経細胞のシナプスに局在することを見出し,そのシナプスにおける機能解析の糸口を得た。その後,酵母ツーハイブリッド法を用いてSept8の結合蛋白質を探索して,シナプス小胞膜蛋白質シナプトブレビンを同定した。シナプトブレビンは細胞内や脳組織内でもSept8と結合していることも見出した。この結果は,Sept8がSNARE複合体形成と神経伝達物質の放出に関与する可能性を強く示唆するものである。2.マウス網膜視神経細胞にSept8が発現している.ことを見出した。また,視神経を薬剤処理してアポトーシスを誘導すると,核内DNAの断片化と平行してSept8の免疫染色性が失われた。これらの結果は,Sept8が神経細胞のアポトーシスのマーカーになることを強く示唆している。3.Sept8類縁蛋白質であるSept14が,胎生後期から新生児期の神経組織に特異的に発現することを確認した。このことは,特定のセプチンが神経発達過程の一時期に重要な機能を有する可能性を示唆する。4.家族性神経痛性筋萎縮症の原因となるSept9(Sept8類縁蛋白質)の1アミノ酸変異が,他のセプチンやRhoのシグナルとの相互作用の異常を惹起することを見出した。この結果は,神経痛の新規治療法の開発のための基礎データになると考えている。
