Research Abstract

本研究では、DNA複製前複合体構築に関与するタンパク質の脱制御によるDNA複製の異常について検討することにより、DNA複製制御機構の一端に迫ることを目標とした。Xenopus卵抽出液中に過剰量のCdt1を添加すると再複製が誘発されるが、同時にDNA複製の抑制反応も誘導されることを示した。この抑制反応はgemininの結合によって阻害されるが、DNA複製前複合体構築反応に必要とされる活性以外の機能によって発揮されるものと考えられた。また、このCdt1の作用はcheckpoint機構のようにDNA複製開始を抑制するものではなぐ、DNA複製時の新生鎖伸長反応を抑制することによることが明らかとなった。これらの結果より、Cdt1のDNA複製前複合体構築への活性をモニターしてDNA複製を抑制する機構が細胞内に用意されていることが示唆された。同様の実験により、Cdc6を卵抽出液に添加した際の影響を検討したところ、やはりDNA複製の顕著な抑制が観察された。このDNA複製の抑制は、CDKによるDNA複製開始活性化以前の過程への効果であること、ATM/ATRを経由するcheckpoint機構を介するものではないことを示唆する結果を得た。このとき、Mcm2-7複合体、Cdk2、Cdc7の両kinaseのクロマチン結合は抑制されなかったが、Mcm4のリン酸化が顕著に抑制された。さらに、このときのCdc7のリン酸化も抑止されていることが観察された。しかしながら、同じくDNA複製開始過程でCdc7の基質となると考えられているMcm2のリン酸化には影響は見られなかった。以上より、Cdc6がCdc7 kinase活性を間接的に阻害することによってDNA複製開始を負に制御している可能性が考えられた。
