Research Abstract

一回膜貫通型サブユニット4F2hc(CD98hc)はLAT1/2とおよびCD98軽鎖等複数のパートナーとヘテロ複合体を形成することが既に知られている。すなわち存在様式であるヘテロタイマー形成そのものが蛋白複合体形成である。 4F2hcがインテグリン等を連結し輸送分子複合体を確立すること、4F2hc過剰発現による不均衡化から組織の過形成が引き起こされ細胞増殖シグナルが発動されること、4F2hcとヘテロタイマーを形成するLAT1/LAT2が胎盤での選択的取り込みについて寄与している。申請者らは現在までに、4F2hcヘテロニ量体型アミノ酸トランスポーターのサブユニットパートナーであるLAT1ノックアウトマウスを作出し、ホモ個体が胎生致死である等のダイマー形成不均衡化に伴う現象を明らかにした。特に母子栄養交換の場である胎盤組織を中心に、正常妊娠過程におけるヘテロ二量体型アミノ酸トランスポーター機能を、LAT1ノックアウトマウスを用いて細胞内シグナル(GTP)伝達について検討した。また、妊娠中期の高血圧症における胎児の未発達病因の一つとして4F2hcを中心とした複合体機能を考慮し、トランスポーターを介した母体-胎児栄養供給と病態機構とインテグリンの発現の有無との関連性を検討した。
