Research Abstract

本研究では、Na/Ca交換体分子複合体の分子構成、制御機構、生理機能および病態との関わりについて解析した。この分子複合体解明の糸口として、α1受容体刺激による血管収縮に1型Na/Ca交換体(NCX1)を介するCa^<2+>流入が関与する実験的証拠を得た。具体的には、NCXl高発現マウスの摘出細動脈ではα1受容体刺激時のCa^<2+>濃度上昇が増大していること、またこの反応は特異的NCX1阻害薬により抑制されることを見いだした。免疫沈降実験から、NCX1は受容体活性化TRPCチャネル・TRPC3およびTRPC6と分子複合体を形成することが示唆された。また、TRPC/NCX1分子複合体の生理機能および病態との関わりを生体レベルで解析するため、NCX1、TRPC3およびTRPC6の高発現マウスおよび低発現マウス(ヘテロ欠損、ドミナントネガティブ変異体導入)を作製した。まず、摘出細動脈のフェニレフリン刺激時のCa^<2+>シグナルを比較したところ、これら3種の高発現マウスではCa^<2+>シグナルの充進、一方、これらの低発現マウスではCa^<2+>シグナルの減弱が観察された。また、NCX1もしくはTRPC3を高発現したマウスではノルエピネフリン投与時に冠動脈スパスム(心電図ST上昇)が誘導されることを見いだした。この反応はNCX1阻害薬投与もしくはTRPC3ドミナントネガティブ変異導入により抑えられた。これらの結果は、TRPC/NCX1分子複合体の機能連関がα1受容体を介する血管トーヌス制御に重要な役割を果すことを強く支持している。
