Research Abstract

オステオポンチンは細胞外基質(マトリックス)蛋白としてまたサイトカインとして機能するシグナルとして骨の制御における重要な役割を担う蛋白と考えられている。本年においてはオステオポンチンのマトリックス蛋白としての機能の解析を特に骨形成シグナルである副甲状腺ホルモンの受容体の制御とのクロストークについて解析した。副甲状腺ホルモンは骨量の増大のみでなく細胞外基質蛋白の相対量を変化させる。この結果、オステオポンチンの存在する場合には副甲状腺ホルモンの構成的変異を持った活性化型のヒト受容体を骨芽細胞で強制発現したマウスが高いレベルの海綿骨量の増加を示す。これに対してオステオポンチンが欠失したバックグラウンドで同じ副甲状腺ホルモンの構成的な活性化変異型の受容体を強制発現した場合には、その既に高いレベルの海綿骨量を更に増加させることが明らかとなった。このことは、オステオポンチンがマトリックス蛋白として機能することを考えた際にこれまでの骨の形質としての発現としての理解を超えるものであり、むしろ逆説的にこの分子が骨形成を抑制することを示すものである。この既に高い骨量レベルに至った副甲状腺ホルモン受容体の活性化変異によるシグナルを超えるオステオポンチンノックアウトによる骨形成は、多数の骨芽細胞の前駆細胞の増加を伴っていた。更に、骨髄における造血系の細胞はむしろ減少し、間葉系の細胞が骨髄を充満する特異的な所見が観察された。全身性の骨形成のパラメーターについてもあるいは局所的な骨形態計測学的なパラメーターのいずれにおいても、副甲状腺ホルモン受容体構成活性型受容体のトランスジェニックマウスを超えるレベルの骨形成が、オステオポンチンノックアウトの共存によって観察されている。細胞レベルにおいてサイクリックAMPレスポンスエレメント(CRE)の活性はPTHの受容体のシグナルによって増加するが、オステオポンチンのsiRNAによる低下によって更に増加することから、そのメカニズムが少なくとも一部は転写であることが推察された。以上の如く、本研究により、細胞外環境因子としてのマトリックス蛋白であるオステオポンチンの新たな機能が明らかとなった。
