Research Abstract

動物細胞のゲノム一次元上、 時間的、 空間的でのプログラムを解明するために、分裂酵母および動物細胞を用いて複製起点の局在や活性化タイミングの解析を行なった。その結果動物細胞において複製タイミングの境界領域を同定し、転写因子、核骨格結合因子であるSATB1がこれに関与することを明らかにした。また複製開始を制御するCdc7キナーゼはMCMのリン酸化を介してCdc45のMCMへの結合を促進することを明らかにした。さらに、Cdc6,ASK,CycliA,CycinB の高発現がES 細胞特有の細胞周期進行を可能にしている可能性を発見し、その発現の制御を担うEmi1を同定した。また分裂酵母を用いそのCdc7ホモログHsk1遺伝子のバイパス変異体の解析から、複製開始を負に制御する因子Mrc1,Bsk2を同定するとともに、複製開始に影響をあたえるその他の要素あるいはクロマチン構造の影響を発見した。これらの発見は複製プログラムの遺伝的制御と可塑性の分子基盤に新しい洞察を与えた。
