| Research Abstract |
我々は新規に構築した多点全反射型蛍光相関分光装置を用いて細胞膜表面におけるタンパク質分子の拡散速度の多点測定を行った。装置は7本の光ファイバーを利用して全反射面におけるエバネッセント面からの7点の蛍光シグナルを受け,蛍光相関測定が可能である。蛍光強度の揺らぎは光電子増倍管で検出を行い,新規にこの研究で構築した相関器で解析した。実際の細胞測定においては膜結合性の蛍光タンパク質(GFP-F)を発現させ,その拡散速度を7点同時に測定を行った。7点各々の拡散速度はほぼ同じであったが,分子数が各点で異なり,細胞膜表面におけるコンパートメントの存在が示唆された。
