Research Abstract

血小板活性化上清が惹起する多彩な細胞応答を明らかにし,その構成成分である生理活性脂質の重要性を明らかにした.中でも,血小板に豊富に含まれ,活性化に伴って放出されるスフィンゴシン1-リン酸(Sph-1-P)に関しては,その詳細な機能的役割を明らかにした.また,血漿,血清中のSph-1-Pの測定の臨床検査医学的応用を進め,その基準値を確定するとともに,慢性肝疾患における低下を明らかにした.
一方,リゾリン脂質としてSph-1-Pと構造上の共通性を有するリゾホスファチジン酸(LPA)に関しては,必ずしも血小板がその産生源ではないことが明らかとなったが,血漿LPA,さらには,血清オートタキシン(ATX:それが有するリゾホスホリパーゼD活性によりLPAが産生される)の測定の臨床検査医学的応用を進めた.その結果,ATXが悪性リンパ腫,肝線維化のバイオマーカーとなることを明らかにするとともに,将来の臨床応用が期待されるATX自動測定の系を確立した.
以上の研究成果は,多機能性細胞である血小板の機能的役割の解明に貢献するとともに,生理活性脂質の臨床検査医学という新しい研究領域の開拓に寄与するものである.
