Research Abstract

本研究は、この小動物の成果を体サイズがヒトに近いミニブタを使い再生医学研究部おける有効性テストシステムを作ることを目的とした。ミニブタを用いて腎臓、小腸および肺の移植実験を行うと共に、胎仔および新生仔の心臓から回収した心筋細胞を用いた再生医療に関する実験を行った。ミニブタの小腸および肺の移植モデルでは、regulatoryT細胞(Tregs)など、移植臓器に対する宿主の拒絶反応に関して前臨床検討した。さらにGFP豚より種々の間葉系幹細胞を樹立し再生医療研究のソースとしての有効性を検討した。樹立された滑膜幹細胞は、励起光下で発光し、関節鏡で観察できることを確認した。幹細胞を使用した関節内組織の再生医療を解析するのに、ブタを用いて、GFPを発光する幹細胞がどのように挙動するか、関節鏡を用いて解析することが可能となった。本システムは、小動物では不可能な、幹細胞の関節内成果での挙動を直接観察することを可能にし、臨床応用での疑問点を明らかにする。
