Research Abstract

大動脈や頸動脈などの中心動脈の伸展性は加齢とともに低下し、心血管疾患の危険因子となる。さらに、加齢に肥満などが加わると動脈伸展性は、より低下する。一方、有酸素性運動の継続は、動脈伸展性を増大させる。しかし、運動が動脈伸展性を増大させるメカニズムは明らかでない。本研究では、血管内皮細胞が産生する一酸化窒素(NO)とエンドセリンに着目して、運動による動脈伸展性の改善メカニズムにNOやエンドセリンが関与しているかを検討した。中高齢者に3ヶ月間の有酸素性運動を実施し、その前後にてNO合成酵素阻害薬およびエンドセリン受容体遮断薬を用いて、動脈伸展性を評価した結果、運動による動脈伸展性の増大には、NOは寄与していないが、エンドセリンは関与している可能性が明らかとなった。また、動脈伸展性が低下している肥満者に3ヶ月間の運動を実施すると、動脈伸展性の増大と血管内皮機能の改善が生じる可能性も示唆された。
