Research Abstract |
一酸化窒素によってDNA中のグアニン(Gua)から生成するオキザニン(Oxa)と発ガンとの関係を検討した。Oxaは遺伝子の修飾を司る酵素(制限酵素、キナーゼ、リガーゼ等)によってGuaと認識され、認識中に架橋反応することはないが、 DNA修復酵素等とは架橋反応を引き起こす。これらの知見と化学合成法を組み合わせてタンパク質-DNA架橋モデルを合成し、この損傷への修復機能の存在を明らかにし、発ガンとの関係が否定できないことを明らかにした。
