Research Abstract

昨年度は、花粉抗原で刺激した患者末梢血単核球のサイトカイン産生プロフィールを網羅的に検討し、IL-17がマーカーとして優れていることを見出した。本年は、マウス脾細胞を用い,TGF-beta+IL-6刺激によりTh17誘導を促進させる評価系を新たに構築した。本系を用いて、ビフィズス菌の菌体成分による免疫抑制メカニズムの解析を行った。その結果、ビフィズス菌成分は、制御性サイトカインであるIL-10やIL-27の産生を上昇させることにより、IL-17産生を効果的に抑制することが明らかとなった。
上記の抗アレルギー機能に関連して、乳酸菌の細胞壁成分であるリポテイコ酸が、腸管バリア障害を保護することによって、腸管におけるアレルゲン侵入を阻止する効果があることも見出した。これは、乳酸菌による抗アレルギー作用の新たな一面であると考えられる。
フラボン類の新たな抗アレルギー作用を評価するため、アトピー性皮膚炎モデルマウスに対するクリシンおよびアピゲニンの摂食試験を行った。その結果、これらフラボンの摂食により、アトピー性皮膚炎の症状悪化を改善する可能性が示唆された。しかしながら、血清中IgEレベルに関しては、アピゲニン摂食群では対照群に対して有意な抑制が認められたが、クリシン摂食群では認められなかった。また、脾臓細胞におけるサイトカインのmRNA発現に関して、クリシン摂食群ではIL-4レベルが、アピゲニン摂食群ではIFN-yレベルがそれぞれ対照群に対して低値を示した。これらの結果より、クリシンとアピゲニンは抗アトピー性皮膚炎作用を示すものの、その作用機序は異なる可能性が示唆された。
