Research Abstract

リンパ球は、高内皮細静脈(HEV)との多段階からなる一連の相互作用を介してリンパ節実質に移行し、免疫応答を開始する。我々はこれまでに、硫酸基転移酵素遺伝子欠損マウスを用いて、第一ステップにおけるL-セレクチンを介したリンパ球のローリングにはユニークな構造を持つ硫酸化糖鎖PNAd(peripheral lymph node addressin)が必須の役割を果たすことを示して来た(Nature Immunology6:1096-1104,2005)。一方、第二ステップでは硫酸化糖鎖の一種であるヘパラン硫酸がHEV上におけるケモカインの提示に関与すると考えられているが、in vivoにおける十分な証明はない。本研究では、新たに樹立するHEV特異的Creリコンビナーゼ発現マウスを用いてHEV特異的に硫酸化糖鎖発現の改変を行い、リンパ球ホーミングの分子機構を解明することを目的とする。本年度は、昨年度樹立した硫酸基転移酵素GlcNAc6ST-2遺伝子のプロモーター/エンハンサーの支配下にCreリコンビナーゼを発現する新規トランスジェニックマウス(GlcNAc6ST-2-Cre Tg)をROSA26レポーターマウスと掛け合わせ詳細な組織学的検討を行い、GlcNAc6ST-2が末梢リンパ節HEVだけでなく、鼻咽頭関連リンパ組織NALTのHEVにも強く発現することを見いだした。硫酸基転移酵素遺伝子欠損マウスを用いた解析を進めたところ、硫酸化糖鎖PNAdが末梢リンパ節のみではなく、NALTへのリンパ球ホーミングにも必須の役割を果たすことが明らかになった。さらに、経鼻的に侵入する抗原に対するアレルギー反応にPNAdが促進的に働くことを見いだした。本研究より、硫酸化糖鎖PNAdが末梢リンパ節および粘膜系リンパ組織の一種であるNALTへのリンパ球ホーミングに関与し、末梢および粘膜リンパ組織の双方において免疫応答に関与することが明らかとなった。
