Research Abstract

我々は、酵母ツーハイブリッド法を用いて成人ヒト心臓cDNAライブラリーから新規エンドセリンA型受容体C末端領域(ETAR C-tail)結合蛋白質としてJab1を単離・同定した。Jab1はETARのユビキチン化、リソソームでの蛋白質分解を亢進させてETARレベルを調節していることを明らかにした。さらに、アゴニストであるET-1でETARを刺激するとアゴニスト誘導性の蛋白質分解が引き起こされ、ETARレベルは時間依存的に減少していく一方、ETARに結合しているJab1量が増加すること、またETARより代謝回転が速いエンドセリンB型受容体 (ETBR)では、ユビキチン化が亢進し、より多くのJab1と結合していることが明らかになった。これらの結果より、Jab1とエンドセリン受容体との結合様式とエンドセリン受容体の蛋白質分解に強い関連性が認められた。
