Research Abstract

我々は骨形成因子の下流因子としてHA4を同定した。HA4ノックアウトマウスは生後の骨量の減少を認め、骨特異的にHA4を過剰発現させたトランスジェニックマウスでは骨量の増加を認めた。このトランスジェニックマウスは卵巣摘出骨粗鬆症モデルマウスの骨量減少を抑制した。これらの結果よりHA4が生後骨量を調節する骨分泌タンパク質である事が明らかとなった。HA4をターゲットとした骨粗鬆症薬の開発のため、HA4受容体の探索も実施した。またHA4は血清中にも検出できることから、血清HA4値を測定し骨疾患の病態進行の指標をするためサンドウィッチELISAシステムを構築した。
