Research Abstract

N-acetyl-aspartyl^glutamate(NAAG)は、哺乳動物の中枢神経系に存在する神経伝達物質の中で3番目に多いものである。NAAGはそれ自体がmGluR3の作動物質であるが、NAAGがNAAG peptidaseにより分解されるとN-acetyl-aspartate(NAA)とglutamateとなる。従って、NAAGは分解されるとNAAGの作用は無くなるが、glutamateが産生されることによりmGluR3だけでなくその他のglutamateの受容体が活性化されることとなる。従ってNAAGを単純に投与することではNAAGの役割を明らかにさせることは出来ない。そこでNAAGの分解酵素であるNAAG peptidase(GCP II及びGCP III)の阻害薬を用いて、glutamateを増加させることなく内因性のNAAGを増加させ、NAAGの侵害刺激伝達に対する役割を検討した。
今回の研究の前に、炎症性疼痛モデル及び神経障害性疼痛モデルにおいてNAAG peptidase inhibitorの静脈内投与・髄腔内投与にて鎮痛効果が見られることを報告している。今回の研究では、NAAG peptidase inhibitorを炎症の局所、もしくは脳室内へ投与することにより炎症性疼痛モデルにおいて良好な鎮痛が得られること見いだした。
NAAG peptidase inhibitorが鎮痛薬として有望なものである可能性が示唆された。
