Research Abstract

平成14~16年度科学研究費海外学術調査"動物の移動・定着に伴う病原体の伝播に関する分子疫学調査"は、1)韓国済州島の済州馬由来分離株の中に日本の木曽馬と同じ病原性プラスミド型(90kb II型)と韓国特有の新しい型(90kb V型)が見いだされ、確かに韓国から日本にこの病原細菌も馬と共に渡来したことが実証された。2)中国の調査では、内蒙古自治区のホロンバイル草原とシリンゴル草原の馬とその飼育環境からロドコッカス・エクイを分離することはできたが、全てが無毒株であった。さらに、3)モンゴル・ウランバートル近郊の遊牧民の馬とその飼育環境からの菌分離を試みたが全く分離されなかった。モンゴルの子馬からロドコッカス・エクイが全く分離できなかったという「真実の意外性」は、病原細菌と宿主(馬)との出会いを再考する一石となった。感染症とは過去のある時代、地球上のどこかで、それまで相互に無縁に近い存在であった生物が偶然生活場所を重ね、その結果、一方は病原微生物に進化し、他方は一方的に害を受ける側の宿主になったものと理解される。その進化した病原微生物がロドコッカス・エクイ強毒株であり、感受性宿主が子馬であると考えていた。本研究では、ウランバートル近郊の子馬からロドコッカス・エクイが存在しないことが、モンゴル全土の馬にも普遍的であるといえるかを明らかにすることであり、スクリーニングとして馬血清中のロドコッカス・エクイ特異抗体の調査、抗体陽性地域の菌分離と病原性更には遺伝子型調査を実施した。驚くべきことに、日本・韓国では全く分離されなかった85-kbI 型プラスミドがモンゴルSelenge 県の馬糞便から分離され、それがオランダの強毒株のパルス型と類似していた。分離された強毒株とオランダ分離株のVap family 遺伝子の塩基配列の解析を行い、この領域の配列が良く保存されていることが明らかとなった。
