Research Abstract

モデル基部植物ヒメツリガネゴケを用いて、被子植物の時計遺伝子のホモログの網羅的な同定・クローニングを行った。それらのうちCCA1/LHYホモログのプロモーターをルシフェラーゼ遺伝子に連結し、コケゲノムに移入し、従来より効果的な発光リズムレポーター株を作出した。このレポーター株をホストとして用いた逆遺伝学的解析により、CCA1/LHYやPRRなどの被子植物の時計遺伝子のホモログの機能を調査し、コケの時計を構成すると思われる遺伝子ネットワークの一部を明らかにし、被子植物の時計機構の分子モデルとの比較を行った。また、コケの色素体シグマ因子遺伝子のひとつPpSIG5の逆遺伝学的解析により、PpSIG5タンパク質がコケ概日システムの出力系の重要な調節因子である可能性を示唆する結果を得た。さらに、コケプロトプラストを用いた、時計遺伝子の日周条件下での発現制御解析のためのトランジェントアッセイ系の開発を行った。
