Research Abstract |
The mtDNA of seven species in this study were showed as circular mapping organization. They contain genes for three rn, for twenty-five trn, for ten nad, for one cob, for three cox, for three atp, for one tat and for three ORF as coding regions in the same way. The organization is identical with that of L. digitata except for nonexistence of ORF 157. The nucleotide length of mtDNA was 37,657bp for L. japonica (Lj), L. religiosa (Lr), L. diabolica (Ld), L. longipedalis (Ll) and was 37,656bp for L. ochotensis. While on the other hand, it was 37,604bp and 37,500bp for L. angustata (La) and L. coriacea (Lc) respectively. As a result of nucleotide alignment, though there was no nucleotide insertion / deletion through total mtDNA among Li, Lr, Ld and Ll, there was one nucleotide insertion / deletion at one of rps 19 between them and Lo. Nucleotide differences are poor among these five species in sum total and pairwise distance (nad6 (partial)-co x1) (: PD partial) between them are 0.02-0.11 through total mtDNA. Nucleotide substitutions and nucleotide insertions / deletions were detected at 69 sites from their package nucleotide alignment, and the value of nucleotide substitution rate was high especially at trnL, trnM, rps19, ORF41 and seven spacer. On the contrary, nucleotide differences were abundant between the five species and the other two species, and PD partial between the five species and La was 4.36-5.85. And nucleotide maxim matching between Li, La and Lc was 95-96%. From this study, genetic differences between Japanese Laminaria species were detected as sequence differences at multi-regions on mitochondrial genome. Within them, several seem to be useful tools for taxonomic and phylogenetic studies. Furthermore, phylogenetic trees for seven species were constructed, and short history for speciation within Lr, Lo, Ld and Ll was presumed.
