Research Abstract |
The small GTPase Arf6 and ARNO are involved in regulation of endocytosis. We demonstrated the crucial role of acidification of early endosomes and V-ATPase/ARNO/Arf6 interactions in the regulation of the degradative pathway during endocytosis. We also found that V-ATPase with the α3 isoform is highly expressed in pancreatic islet, and localized to membranes of insulin-containing secretory granules in b cells. oc/oc mice, which have a null mutation at the α3 locus, exhibited a reduced level of insulin in the blood, even with high glucose administration. These results suggest that the α3 isoform of V-ATPase has regulatory functions in ,exocytosis of insulin secretion.
