Research Abstract

シェーグレン症候群(Sjogren's syndrome ; SS)モデルIQIマウスにおけるB細胞優勢の下顎腺炎(B細胞性下顎腺炎)を遺伝学的に解析した。その結果、B細胞性下顎腺炎の量的形質遺伝子座(QTL)は少なくとも3個有り、単独で効果を発揮する主要QTL(autoimmune sialitis in IQImice, associated locus 1 ; Asq1)が第6染色体前端に、Asq1の効果を増強するエピスタシスQTLが第3染色体後方(Asq2)に、また、逆にAsq1の効果を抑制する抵抗性遺伝子座が第1染色体後方(Asq3)に位置することを明らかになった。Asq1の1LOD区間内に位置するIca1に注目し、そのcDNA塩基配列を対照C57BL/6(B6)と比較した結果、IQI-B6間で6個の一塩基多型があり、2個のアミノ酸多型が生じていた。また、ウエスタンブロット法では、IQエマウスの血清中に抗Ica1自己抗体の存在を示すバンドカミ検出された。以上の結果より、IQIマウスにおいて、Ica1に対するB細胞自己免疫が成立していることが明らかになり、原因として、1ca1の塩基多型の可能性が挙げられた。
