Research Abstract

種々の型のターピリジン結合ヌクレオシド誘導体の合成を行ない、これらを用いてターピリジン・Cu(II)錯体残基を鎖中央に2個含む2'-O-メチルRNA オリゴマーの基本形4種を構築した。これらの人工RNase は相補的配列を含む短鎖RNA や長鎖RNA を位置特異的に、また短鎖RNA については高効率的に切断することが分かった。また天然酵素のように触媒回転能を有していた。加えて、ほとんどの酵素の活性は、以前に筆者が開発した同型の酵素よりも高いものであった。一方、RNA 変異体基質を用いた切断反応の解析により、人工酵素の構造・活性相関について重要な知見を得た。
