Research Abstract |
1. Matrix metalloproteinase (MMP) expression The mRNA expression of MMP-2 and MMP-9 was shown to decrease by using RT-PCR in the uteri of 10-day-old mice exposed to diethylstilbestrol neonatally (DES-mice)than those in vehicle-treated control mice. Immunohistochemical study demonstrated that MMP-2 expression significantly decreased in the uteri of DES-mice. In situ zymography using fluorescence-labeled gelatin, enzyme activity of MMP-2 was not detected in the uteri of both experimental groups, although granulocytes showing MMP-9 enzyme activity increased in the DES-mice uteri. 2. Tissue inhibitor of metalloproteinase (TIMP)expression RT-PCR showed that no significant differences in the mRNA levels of TIMP-1, -2, -3 and -4 between the control and DES-mice. 3. Collagen expression The mRNA expression of collagen type-I, -III and -IV was shown to be almost the same in uteri of the control and DES-mice by using RT-PCR. 4. Vascular endothelial growth factor (VEGF) expression Immunohistoichemical study showed that VEGF expression in the uterine stromal cells was much lower level than in those of controls.
