Research Abstract

リポ多糖類の静脈内投与や終板器官周囲部へのプロスタグランジンE2(PGE2)注入による発熱は視索前野にGABAA受容体拮抗薬を前投与しておくと阻止された。GABA拮抗薬によって視索前野の発熱抑制作用が増強されたためと考えられる。しかし、GABA拮抗薬の投与は片側で有効であり、PGE2投与部位とは同側でも対側でも効果に変わりがなかった。この結果から終板器官周囲部のPGE2感受性ニューロンが視索前野に投射して発熱反応を引き起こすという仮説は否定され、視索前野から終板器官周囲部のPGE2感受性機構に影響する可能性が想定された。次に、PGE2感受性の発熱誘起部位におけるGABA受容体の役割を調べた。この部位にGABA拮抗薬を注入するとPGE2と同様の反応が誘起された。GABAA受容体作動薬ムシモールを前投与しておくとPGE2投与によって誘起される反応が全て阻止された。この部位のGABA感受性の熱産生抑制機構が抑制されることがPGE2発熱の機構の少なくとも一部である可能性が考えられた。
