Research Abstract |
This research has been focused on establishment of in vivo imaging system to examine the process of muscle regeneration. Three different approach has been taken. First, using LacZ transgenic mice, I prepared to integrate fluorescent microscope system. With this system, We succeeded to detect terminal Schwann cells on skeletal muscle in ligving animal. We tried to observe regeneration of sternomastoid muscle, cremaster muscle, gracilis muscle, latissimus dorsi muscle and flexor hallucis brevis. We also screened fluorescent markers and methods of muscle injury. Second, using Zebrafish embryo, we traced the muscle precurcer cells by confocal laser microscopy. Third, we made and screened monoclonal antibodies to detect subpopulation of muscle precurcer cells at specific stage. Then we could identify a couple of novel membrane proteins. Based on the data above, we keep trying to establish in vivo imaging system to examine the process of muscle regeneration.
