Research Abstract

(1) XDH阻害剤複合体のデータ収集、精密化
ウシXDHと国内で開発した数種類の阻害剤との複合体の構造解析を行なった。今年度は天然に存在する不活性型酵素であるデサルフォ型酵素との複合体につきデータ収集、精密化を行い、完了した。生体内ではデサルフォ型酵素はXDHの総量の20~30%を占めており、活性中心モリブデンに配位している硫黄原子のひとつが酸素に置き換わっている。デサルフォ型酵素が活性型にコンバートされることにより活性調節を行う機構も考えられている。デサルフォ型酵素と各種阻害剤の構造を解明することは阻害剤の阻害機構解明、酵素反応機構解明のための重要な知見となる。
(2) 構造ベース創薬の試み
上記研究の知見に基づき、酵素と安定な反応中間体を形成し酵素反応を強力に阻害する化合物を国内製薬会社と共同で設計した。同化合物とXDHとの複合体結晶構造を作成中である。
(3) ラットXDHおよび変異酵素の結晶化、データ収集、精密化。
哺乳類XDHは構造変化を起こすことで活性酸素生成の制御をおこなっていることをラット酵素蛋白質の変異体についてX線結晶解析により構造解析を行い明らかにした。これらの研究は本酵素蛋白質が乳汁分泌に係わることを意味し、系統発生における蛋白質の使い分けという興味ある課題である。本プロジェクトの推進により得られた知見により、活性変換にかかわる新たなファクターとして酵素タンパクのC末端が関与していることが予想された。今年度はC末端欠失変異酵素を作成、結晶化し、データ収集を行った。現在精密化作業を行っている。
