Research Abstract

肺癌に於いて、ヒトの個性(p53 とMDM2 の多型)とがんの個性(組織型や遺伝子異常)が、発がん、生物学的動態、治療に対する反応性等に与える影響を検討した。その結果、(1)一部の腺癌の発生にはMDM2 の過剰発現が関係しているが、その過剰発現は多型と関係がない、(2)腺癌の各組織型はそれぞれ特有の遺伝子異常を有し、形態から遺伝子異常を推測することが可能、(3)P53, K-ras, EGFR, p16 遺伝子のうち、前2者がともに異常を有する場合に最も予後が悪い、(4) p53 多型と治療反応の関係は更なる検討が必要、であった。以上の結果は、分指標的治療薬開発の標的分子を、また、がんの個性に応じた治療法選択の必要性を、示唆するものである。
