Research Abstract |
Recently, by completing genome sequencing, we revealed that Vibrio parahaemolyticus a causative agent of acute gastroenteritis to humans, posseses two sets of genes for the type III secretion system, TISS1 and TISS2. In this study, we analyzed the conditions and mechanisms of gene regulation for the type III secretion systems in V. parahaemolyticus. Expression of the genes for the type III secretion system components and effectors in various in vitro conditions revealed that the TISS1 genes are mainly expressed in a log phase, while the TTSS2 genes are in late log and stationary phases. Both the TTSS1 and TSS2 are more expressed in culture with 1% or less NaCl than 3% NaCl. Also, both systems are preferentially expressed at the temperature higher than 30℃. There results suggested the appropriate conditions for the expression of the type III secretion systems. We identified the regulator genes for the TTSS1 and TTSS2 genes, respectively. The one for the TTSS2 regulates not only the genes for TTSS2, but also almost all genes present on the pathogenicity island Vp-PAI. Furthermore, by the analysis using microanay, we demonstrated that the TTSS2 regulator does not affect the gene expression of the gene outside the Vp-PAI, thus the regulator is suggested to be specific to the gene on Vp-PAI. These results provide an insight into the TTSS gene regulation in V. parahaemolyticus.
