Research Abstract

培養肝癌細胞はHCV複製が可能な細胞である。本研究ではHuh-7細胞株からHCVレプリコン複製能の高い細胞クローンを樹立した。これら複製能の高い細胞クローンではアポリポ蛋白質遺伝子の発現亢進が認められた。これらの遺伝子をノックダウンするとHCVの複製が抑制された。また、複製効率の良い全長HCVを作製するにための複製効率の良いHCVサブゲノムレプリコンを樹立できた。一方、その過程において欠損ゲノムを有するHCVを同定するとともにその感染性をin vitroで証明できた。
