Research Abstract

前年度に引き続き、慢性炎症が関与する癌とその発生母地となる各組織においてミトコンドリアDNAを系統的に解析し、発がんの分子機構を明らかにするとともに発がんにおけるミトコンドリアゲノムデータベースを構築の輪として以下の項目を推進した。
1)種々の炎症性発癌におけるミトコンドリアゲノム解析:口腔粘膜癌、肺癌、胃癌、肝癌、大腸癌において、ミトコンドリアゲノムを解析して変異を同定した。
2)ミトコンドリアゲノム変異と臨床経過の相関の解析:同定したミトコンドリアDNA変異情報において臨床経過との関係を解析し、有意な相関を認めた。
3)ミトコンドリアゲノム解析による段階的発がんの評価:「本研究で各臨床症例から得たミトコンドリアDNA変異と臨床経過との相関」を「活性酸素種暴露細胞}におけるミトコンドリアDNA変異蓄積の変遷と形質転換の様相」に照合し、その連関を解析した。
4)発がんにおけるミトコンドリアゲノム変異の基礎医学的解析
これらの解析により、臨床症例のミトコンドリアDNA変異と発がんとの関係の統計学的処理を推進した。本解析にて得られた結果と基礎実験データから、ミトコンドリアDNA変異を基準とする発がん危険度を理論に基づいて数値化することを試みた。さらに、血液生化学データおよび組織病理学的評価とミトコンドリアDNA変異を総合的に解析することにより、発がん危険群としての炎症病態の新たな評価基準の構築を推進した。以上のことより、ミトコンドリアDNA変異と発がんとの直接の因果関係を明らかにすべく、研究を統括した。
