Research Abstract

本研究はカドミウム(Cd)と環境化学物質の複合影響を疫学的・実験的に明らかにすることを目的とする。疫学的な調査研究により、Cd汚染地域の住民では、尿中にvitamin-D binding proteinが排出されており、その結果vitamin Dの代謝に異常が生じ、骨障害に通じる可能性を明らかにした。またCdと環境化学物質の複合影響を評価するためのin house cDNAマイクロアレイの構築とその検証を行った。本研究において作成したアレイは、目的特化型であり、安価でかつ後の解析が容易なものである。そのために、トランスクリプトーム情報をもとにして、ストレス応答遺伝子、アポトーシス関連遺伝子、生体異物の代謝に関与する核内受容体の応答遺伝子等の収集を行い、マイクロアレイの構築を行った。代表的な環境化学の急性投与、および長期投与における遺伝子発現の変動を解析し、構築したアレイの有用性を明らかにした。さらにクラスター解析とパスウェイ解析を行い、これまでの報告と合致する成績を取得するとともに、化学物質投与により影響を受ける新規パスウェイを明らかにした。
