Research Abstract |
1) Osteoprotegerin (OPG) knock out (KO) mice was employed as a model of bone metabolic disorder and vascular dysfunction. The normal phosphorus diet (phosphorus 0.6%) and the high phosphorus diet (phosphorus 1.2%) were supplied to the model mice. As a result, the control of the serum FGF-23 less elevated reaction was clarified in OPG KO mice by the phosphorus loading. It was cleared that OPG derived osteoblast mainly control of the production and secretion of Fibroblast Growth Factor-23 (FGF-23). 2) The secondary hyperthyroidism model was adopting established as an abnormal phosphorous metabolism that progressed along with the renal damage. Secondary hyperparathyroidism model rats were induced high phosphate diet in 5/6 nephrectomized rat. As a result, the place of FGF-23 production induced high phosphate diet was bone tissue. 1, 25 (OH) 2D3 as a active form of vitamin D was able to induce the skeletal FGF-23 elevation but not parathyroid hormone. 3) The organ culture method was tried to examine a direct effect of phosphate on rat vessel as an established and successful methods. The aortic vascular ectopic calcification was induced high phosphorus culture in vitro condition. In addition, sodium-phosphate co-transporter certainly works some role in this vascular calcification mechanism.
