Research Abstract |
Since both preclinical and clinical studies have clearly implicated the serotonin (5-HT) and norepine phrine neurotransmitter systems in the mechanism of action of antidepressants, a monoaminergic hypothesis of the pathophysiology of depression was put forward. Our study investigated how SSRI, SNRI acted in vivo neuron, and which protein bond antidepressants. We used milnacipran (SNRI) hiaffinity colum and applied rat brain cytosol fraction. Antidepressants binding protein was detected the 2D analysis system variable imageanalyzer, and identify by the mass spectrometry. Milnacipran bond 4-Fluoro-7-nitro-2, 1, 3- benzoxadiazole (NBD-F) was detected in vivo PC12 cells by confocal microscope. We investigated that milnacipran bond NBD-F colocalize antidepressants binding protein by confocal microscope.
