Research Abstract |
1. Characterization of PGHAM-1-Luc cells in vitro We developed a new cells named PGHAM-1-Luc which is special PGHAM-1 cells with luciferase activity transduced by AAV vector. PGHAM-1-Luc-1 has most activity of luciferase among five subclones from PGHAM-1-Luc cells. Luciferase activity is measured by IVIS Imaging System (IVIS 100, Xenogen) with luciferin. Doubling time of PGHAM-1-Luc-1 was 18.2 hours that is longer than that of original PGHAM-1. 2. In vivo transplantation experiment using PGHAM-1-Luc-1 cells Three models for PGHAM-1 ; 1)primary pancreatic cancer and simultaneous liver metastasis by intrapancreatic transplantation, 2)liver metastasis alone by intrasplenic transplantation, 3)peritoneal dissemination by intraperitoneal transplantation, within twenty-one days after transplantation. Results of transplantation of PGHAM-1-Luc in hamsters. Intrapancreatic transplantation model : Bioluminescence image was shown in primary and metastatic tumors at 14-21 days. The sin of bioluminescence was almost same as actual size at autopsy. Intrasplenic transplantation model : Bioluminescence was found in spleen without induction of liver metastasis. Intraperitoneal transplantation model : Very low number of disseminated nodules on peritoneum with faint bioluminescence. From above data, the metastatic activity of PGHAM-1-Luc-1 was less than that of PGHAM-1. 3. Pathology of tumors Histopathology of pancreatic tumor by PGHAM-1-Luc-1 transplantation is moderately of poorly differentiated adenocarcinoma. PGHAM-1 is well-differentiated one. Any changes would be induced through gene induction.
