| Research Abstract |
アミノ酸トランスポーター機能抑制を目的とするsystem L : LAT1/4F2hc標的治療は有力な悪性glioma 治療戦略の一つと期待される.System L阻害剤であるBCH治療により, glioma 細胞死は亢進したが, PTEN 遺伝子異常をもつglioma 細胞では, 生存に関与する内因性シグナル分子Akt のリン酸化はむしろ亢進し, paradoxical な変化が認められた.siRNA による選択的LAT1機能阻害を目的にLAT1 siRNA 発現ベクターを作製し, LAT1 蛋白の発現が低下したglioma 株を分離樹立した.ヌードマウス皮下腫瘍形成能実験にて本細胞の造腫瘍性の低下が認められた.
