Research Abstract |
It has been supposed that the immune system in the inner ear is involved in the inner ear disorders. However, the details are unkown. And the most of molecular mechanisms in the immune system of the inner ear are not clarified. CD47 is a transmembrane protein which associates with integrin. CD47 and its ligand SHPS-1 constitute a cell-cell communication system through each extracellular region. This CD47-SHPS-1 system has been shown to participate in regulation of a variety of biological functions. The interaction of CD47 with SHPS-1 has been studied in the immune system. In migration of macrophages and neutrophils into epithelium with inflammation, it is supposed that SHPS-1 expressed on neutrophils interact with CD47 localized between epithelial cells and migration of neutrophil is promoted. The aim of this study is to clear physiological roles of CD47 in the immune system of the inner ear and the inner ear disorders by showing expression and localization of CD47 in the inner ear and participation of CD47 in the interaction of neutrophils with epithelial cells of endolymphatic sac, which plays a central role in the inner ear immune system. To investigate expression and localization of CD47 and SHPS-1, inner inners were removed from mice and frozen sections of cochlea, vestibule and endolymphatic sac were prepared. Immunohistochemical studies have been performed by using anti-CD47 antibody and anti-SHPS-1 antibody. Clear results have not been obtained yet. Now studies are continued. Additionally analysis of the morphology of the inner ear in CD47 knock-out mouse and SHPS-1 knock-out mouse is planned.
