Research Abstract

生体内耳のバイオプシーは不可能なので、ヒト内耳疾患の病態はいまだ不明の点が多く、肝や腎など生体バイオプシーが出来る臓器の疾患と比較して決定的にその解明が遅れている。そこで生体内耳の変化を「蛋白病理学的」に把握できる唯一のサンプルである外リンパに着目した。
本研究では、Ettan DIGEシステム(高感度で定量的な2次元電気泳動蛋白解析装置)を用いて外リンパ蛋白の網羅的ディファレンシャル解析を行い、難聴患者の内耳蛋白の変化・変性の本態を探り、治療のターゲットとする蛋白を同定する事を目的とした。
Ettan DIGEシステム等を可動させ、基礎実験を行いながら至適条件設定を行い解析を行った。その結果、現在の所我々がすでに発見し、臨床応用しているCTP蛋白以外の有効な内耳特異的マーカーは発見されていない。この原因として、生命現象にかかわる重要な蛋白で、微量にしか発現していない物を検出するには感度が十分ではない可能性が指摘されている。現在研究を続行中である。 交付額
