In this study, we examined molecules which are increasing their gene expression in peripheral T cells of mice developing experimental autoimmune uveoretinitis (EAU) by immunization with interphotoreceptor retinoid-binding protein (IRBP). As the results, mRNA expression which increased more than 5 fold on both 7 and 14 days after immunization were cytokines associated with Th17 cells except for those of Thl cells, implicating that Th17 cells were also involved in the occurrence of EAU. Subsequently, we collected peripheral T cells from behcet's patient with uveitis, stimulated them with S-Ag and IRBP, and measured production of IFN-g, IL-17, IL-6, TNFa, and IL-10. Unexpectedly, positive rates of T cells reactive for S-Ag and IRBP were statistically not different between Behcet's patients and healthy controls. However, the amounts of several produced cytokines were more in Behcet's patients than in healthy controls, in which Behcet's patients produced more IFN-g, IL-17, and IL-6 by stimulation with IRBP, and more IFN-g and TNFa by stimulation with S-Ag than those of healthy controls. We continue this series of experiments, also using peripheral T cells obtained from Behcet's patients who have not had uveitis, sarcoidosis patients, and VKH patients, et. al.
