Research Abstract

目的1) in vivo研究としてヒト歯肉溝滲出液(GCF)中ならびに顎関節症滑液中の骨代謝関連因子の探求、とくにRANKLとの関連性が強い破骨細胞由来Cathepsin-Kの定量2)in vitro病態モデル(炎症性サイトカイン処理マウス骨芽細胞)により、惹起されるRANKLならびにアポトーシスを誘導するcaspaseを中心としたシグナルカスケードの分子機構の解明3)病態モデル(炎症性サイトカイン処理マウス骨芽細胞ならびにストローマ細胞)により、惹起されるOPG産生抑制シグナルカスケードの分子機構の解明4)骨芽細胞の分化・増殖におけるosteoprotegerin(OPG)の新規な生理的役割について詳細な基礎的な検討を行い、新たな歯周関連疾患治療戦略の構築をめざすことを目的とした。
結果1) 健常者、顎関節症のdisk displacement with reduction, disk displacement without reduction, osteoarthritisの4群から滑液を得てRANKL, OPG, RANKL/OPG ratioを測定した。ヒト顎関節症、とくにosteoarthritis患者滑液中にRANKLが存在し、OPG量が健常者と比較して統計的有差を持って低下を示し、有意にRANKL/OPG ratioが亢進することを初めて明示した。Osteoarthritis患者滑液がin vitroでヒト末梢単球からの破骨細胞形成ならびに活性化能を持つことを確認し、Osteoarthritisの骨破壊の機序にRANKL/OPGが関わる可能性を初めて明示した(Wakita, Mogi et al., J. Dent. Res.)
2) 健常者GCFとの比較から、ヒト歯肉溝滲出液GCF中の破骨細胞由来と考えられるプロテアーゼCathepsin-Kと破骨細胞分化誘導因子RANKL濃度が歯周病の進行・ステージにより上昇変動が見られ、両者とも統計的有意な増加ならびに相互に相関を認めた(Mogi, M. et al., Archs Oral Biol., 2007)。
3) マウス骨芽細胞様細胞MC3T3-E1細胞において, 炎症性サイトカイン誘導アポトーシスシグナルは, mitogen-activated protein(MAP)キナーゼの活性化を伴うことを明らかに、iNOS依存性アポトーシス細胞死システムの誘導を調節する上でp38MAPキナーゼカスケードが, 重要な役割を担うことを明示した(Kuzushima, Mogi et al., Archs Oral Biol).
4) OPG-knockout mouseにおける血中RANKLおよびOPGの変化と骨代謝異常の関連性について精査し、OPG-knockout mouse血中RANKL濃度の顕著な増大を発見した。本実験結果はOPGの新規な生理的役割として膜結合型RANKLの可溶性型RANKLへの移行を調節するユニークな作用を持つことを初めて明示した(Nakamichi et al., J. Immunol., 2007)。
5) ヒトgingivitis患者歯肉溝滲出液(GCF)中のサイトカインの変動を健常者GCFサンプルとの比較探索を行ない、EGF superfamily のtransformng growth factor-αが特異的にgingivitis群において低下している事を明らかにした(Mogi et al., J. Immunoassay Immunochem., 2009)
