Research Abstract |
In 2006, we examined preventive effect of folic acid for dioxin (TCDD) induced cleft palate in vivo. ICR strain mice of 8-10 weeks of age were used. After 2 weeks administration of folic acid. female mice were impregnated. TCDD was administered with gastric tubes at 40 μg/kg at gestation day (GD) 12, and all fetuses were examined at GD 18. However, folic acid had no preventive effects on TCDD induced cleft palate. Then, in 2007, we changed the aim of this research to investigation of the more detailed mechanisms of TCDD induced cleft palate. In the result of SEM (scanning electron microscope), HE staining, immunohistochemical staining (TGFB3, MSX1, FGFR1/2, LHX8), and organ culture examination, development of fetal palates administered TCDD delayed, but fused as same as normal palates once, however, after fusion phase they ruptured and became cleft palates.
