Research Abstract |
To find novel sleep regulating materials, traditional medicines used for the treatment of insomnia were screened by locomotor activity assay, the followed by sleep assay system. In the present study, we used the assay method to evaluate the sedative effect of herb extracts of Hemerocallis fulvar. sempervirens a hypnotic medicine in Okinawa prefecture of Japan, and the seed of Zizyphusjujuba MILL., a Chinese natural medicine Zizyphi Spinosi Semen after oral administration in mice. The sedative effect in the mice treated with water extract of roots of the Hemerocallis genus was observed, and less effect was shown with water extracts that of leaves and flowers. Oxypinnatanine, an constituent of the water extract of roots of Hems/veal/is genus, was found to exhibit the sedative effect in mice. EtOH/Water extract of Zizyphi Spinosi Semen showed the sedative effect in mice, but its efficacy was relatively weak. Jujuboside A, the isolated compound from Zizyphi Spinosi Semen, was also somewhat found to possess the less sedative effect. We are now evaluating the somnogenic effect of these compounds by electroencephalogram and electromyogram sleep bioassay system. These compounds may provide a potential therapeutic approach for insomnia
