Research Abstract

研究2年目として脳内腫瘍のイメージングを行った。GFPトランスジェニックマウスの大腿骨から骨髄細胞を採取し、正常C57BL/6マウスに移植した。1週間後、株化グリオーマ細胞GL261を定位的に右脳の線条体へ局所注入した。個体レベルでの脳内の変化を見るために、経時的に小動物用Magnetic Resonance Imaging (MRI)や液晶同調フィルターが装備された蛍光in vivo imaging装置を用いて、脳腫瘍の増殖やGFP発現細胞の動態を撮像した。腫瘍移植後3日、1週、2週、3週においてマウスをPBSで灌流し、脳を採取した。採取した脳から凍結切片を作製し、免疫組織化学的解析を行った。また、脳切片の一部からtotal RNAを抽出しreal time PCRによる解析を行った。MRI撮像より、腫瘍移植後2週の時点から腫瘍の増殖が確認された。2週目と3週目の骨髄移植を行ったマウスと行わなかったマウスを比較すると、骨髄移植を行ったマウスにおいて腫瘍の大きさが有為に増大していることがわかった。脳内におけるGFP発現細胞の動態について調べてみると、腫瘍移植後から2週にかけて腫瘍細胞注入部位へ徐々に集積し、2週をピークに減少していくことがわかった。腫瘍移植前に脳内で見られるGFP発現細胞のほとんどは、未分化骨髄細胞マーカーの一つであるER-MP12抗原を発現していたが、腫瘍移植後2週目の脳内に見られるGFP発現細胞ではその割合が減少し腫瘍巣及びその周囲の細胞はミクログリアやマクロファージのマーカーの一つであるCD11bを発現していた。また、骨髄由来細胞やミクログリアには発現が見られGL261では発現が見られないカテプシンSの発現が顕著に上昇していた。蛍光in vivo imagingによって脳内カテプシン酵素活性を検出すると、腫瘍移植後2週にかけて上昇していることがわかった。
