Research Abstract

本研究は培養細胞およびマウス個体に高エネルギー・高LETの重粒子線を照射し、飛跡に沿ってDNA二本鎖切断(DSB)を生成させる。この直接的な放射線によるDSBとバイスタンダー効果によって近傍の細胞に生成するDSBに集まるDSB損傷認識タンパク質の照射後の経過時間毎の挙動を明らかにすることを目的としている。
高エネルギー重粒子線の飛跡に集まるDSB損傷認識タンパク質の挙動を明らかにするため、昨年度に引き続き、我々は重粒子線(Fe,500MeV/u,200keV/μm)照射後のこれらのタンパク質のフォーカス形成とリン酸化を免疫化学染色法とフローサイトメトリー法を用いて解析した。リン酸化ヒストンH2AXのフォーカス形成は放射線によるDSB損傷部位で起こることがよく知られているので、我々は線量依存的な飛跡の検出に用いた。
その結果、理論値とよく一致して線量と飛跡の数が一致していた。また、X線と同様に重粒子線の線量が増えるのに従ってヒストンH2AXのリン酸化量は増加したが、X線に比べて重粒子線照射後のヒストンH2AXのリン酸化残存量が多いことを明らかにした。このことは重粒子線による損傷が修復しにくいことを示しているのかも知れない。さらに、我々はDSB損傷認識タンパク質(特に放射線照射後にリン酸化したATMセリン1981、ヒストンH2AXセリン139、Nbs1セリン343、Chk2スレオニン68、DNA-PKcsスレオニン2609)の重粒子線照射後の挙動を調べ、リン酸化ヒストンH2AXのフォーカス形成部位とよく一致していることを明らかにした。
高エネルギー・高LETの重粒子線はX線やγ線などの低LET放射線やその他の化学物質などと異なり、その飛跡に沿ってDSBを生成することから、DSB認識機構の解明にとって、有用なツールと考える。
