低酸素環境におけるマンガンのバイオミネラリゼーション

Research Abstract

嫌気条件下で、マンガン酸化真菌Paraconiothyrium sp.WL-2株を通常の培地に硝酸イオンを添加して培養したところ、嫌気中では、25日間たっても全くMn^<2+>の酸化が認められなかった。培養時間に応じて、嫌気雰囲気下で真菌と培養液を孔径0.22μmのメンブランフィルターにてろか分離し、ろ液を大気中で保管したところ、5日後に黒い沈殿を形成し、ろ液のMn度分析から、Mn^<2+>が酸化されていることが確認された。培地のpHは6.9±0.1であった。このpH領域ではMn^<2+>の化学的酸化は起こり得ない。すなわち、このマンガン酸化真菌Paraconiothyrium sp.WL-2株は嫌気条件下であっても、マンガン酸化酵素(マンガンペルオキシダーゼ)を産生することが明らかとなった。また、マンガン酸化酵素の産生と硝酸イオンの消費は関係が認められなかった。このことから、好気条件においてみられるParaconiothyrium sp.WL-2株のMn^<2+>の酸化プロセスを推定すると、マンガン酸化酵素(マンガンペルオキシダーゼ)の産生と酸素を使った酵素反応によって成り立っていると考えられる。嫌気的環境においてもこのマンガン酸化真菌Paraconiothyrium sp.WL-2株は酵素を産生し、それらが準表層環境に曝されたときに、酵素反応により鉱化作用が始まり、準表層環境におけるバイオレメディエーションに寄与すると考えられる
